Pico discovers that he is trapped in a time loop while singing against BF. He eventually determines that GF is the reason the resets continue and kills her. After doing so, he experiences an immense surge of power and gains EXP. He then pursues BF, although BF escapes by falling into the Underground. Pico continues killing more people and becomes practically immortal by the time of The Uprising. His story demonstrates how the repeated-reset concept can push familiar characters toward increasingly extreme actions.
Sans is normally known as a lazy but beloved skeleton with a talent for jokes. In this story, however, he carries knowledge that no one should have to bear. After enough resets, Sans begins remembering more than he should. He realizes that the human's repeated actions cannot be stopped through ordinary means. His conclusion is devastating: he needs enough EXP to make it impossible for the human to win. Sans is also Papyrus' brother, making his decisions even more painful because of what eventually happens to his own kind.
Papyrus remains a jovial character with a love of puzzles and pasta. He dreams of becoming a Royal Guard and gaining enough popularity to have many friends. His personality contrasts sharply with the darker circumstances surrounding him. That contrast makes the events involving Papyrus particularly significant to the overall story.
Phantom Papyrus is described as a creation of Sans' mind, brought about by the guilt he feels over killing his brother. However, the phantom still appears to contain part of Papyrus himself. This character adds a psychological element to the story, representing the emotional consequences of Sans' choices rather than simply serving as another opponent.
Undyne is the fierce leader of the Underground's Royal Guard and a symbol of hope for its inhabitants. Even after she should have been defeated permanently, she continues fighting because she believes she may be the only force capable of stopping the murderers before they destroy everyone and ultimately threaten the world. Her determination makes her one of the game's most important figures.
Mettaton NEO appears after the CORE is destroyed due to a lack of maintenance. Forced to transform into the NEO form while submerged in lava, Mettaton adapts to the extreme environment and emerges with a distinctly "hot" new appearance. The unusual circumstances surrounding this transformation contribute to the game's strange and unpredictable cast.
Sergeant John "Tankman" Captain is another character you encounter in the game. He is the last remaining member of his army after Pico kills the others. His presence reinforces the destructive scale of Pico's actions and adds another survivor to the game's increasingly bleak world.
